Description of the Service Centre
The BioLab is a Biological and Chemical Analysis Facility at FCT-NOVA. This facility is devoted to the comprehension of cellular and molecular mechanisms, from the in vitro characterization of molecular interactions to the in vivo validation in model animals. The BioLab facility gathers in a single unit a unique set of state-of-the-art biophysical and biological technologies. Circular Dichroism

Circular Dichroism (CD) is an absorption spectroscopy method based on the differential absorption of left and right circularly polarized light. CD spectroscopy can provide information on the structure of proteins, peptides, nucleic acids and other chiral compounds having a wide range of applications in many different fields. Application examples: Proteins - Secondary structure prediction; Tertiary structure fingerprint; Detection of conformational changes; Structural analysis of recombinant native proteins and its mutants; Effect of experimental conditions on protein conformation; Measurement of protein stability; Follow-up of protein folding; Studing of bindining interactions. Nucleic Acids - Determination of nucleic acids conformations.

Differential Scanning Calorimetry (DSC) is a thermoanalytical technique that measures the heat absorbed or released by a sample as it is heated or cooled. DSC is widely used for the thermodynamic analysis of biomolecules and nanoparticles. Application examples: Protein stability and folding; Binding interactions: Protein-ligand, Protein-Protein/Lipid, Drug-Lipid or Drug/protein-DNA; Characterization of membranes, lipids, nucleic acids and micellar systems.

MicroScale Thermophoresis (MST) is based on the directed movement of molecules in a temperature gradient which strongly depends on a variety of molecular properties such as size, charge, hydratation shell or conformation thus being a powerfull technique to quantify biomolecular interactions.Application examples: Protein folding; Thermodynamics; Interactions in complex liquids; KD determination; Ultra-high affinity measurements; Enzyme kinetics.

Multi Parametric Surface Plasmon Resonance

Traditional surface plasmon resonance is an optical technique established as a powerful method to monitor label-free biomolecular interactions in liquids. At BioLab the Bionavis™ SPR Navi 200 is available as an improved and versatile system, that uses an increased angular scan range to produce a complete SPR curve (multi-parametric SPR) enabling new insights into interactions. This increased angular range also allows the characterization of the interactions of surfaces with analytes, with molecular weights ranging from a few Da to several KDa, in the gas or liquid phases. MP-SPR has multiple applications both in Life Sciences and Material Sciences. It allows the accurate measurement of molecular interactions (e.g. kinetic and affinity constants) and determination of layer properties (e.g. refractive index, thickness) with the help of the softwares TraceDrawer and LayerSolver, respectively.

We also operate with SX20 Stopped Flow accessory that allows CD and absorbance stopped-flow measurements of high sensitivity. The Pro-KIV Kinetic Global Analysis and the Global 3 Thermal Analysis software enable the analysis of multi-wavelength kinetics and temperature ramps respectively.

The Attune™ NxT Acoustic Focusing Cytometer is a 2-laser (red and blue), 6-colours system that uses ultrasonic waves in combination with hydrodynamic forces, to position cells into a single, focused line along the central axis of a capillary allowing the collection of more photons for high-precision analysis at superior volumetric sample throughput. This highly sensitive system is designed to acquire data at up to 35000 events/second, detect particles with diameters from 0.5 µm to 50 µm and rare events at sample rates up to 1000 µL/min. The Attune™ NxT Software is designed to provide powerful data acquisition and analysis and can perform pre- and post-compensation (automated or manual).

The Nano Temper technologies Monolith NT.115 measures biomolecular interactions via detection of fluorescent dyes or fluorescent fusion proteins, providing a broad application range to detect interactions between any type of biomolecules and for pM to mM binding affinities. The MO.Affinity Analysis software allows for a straightforward analysis and evaluation of the MicroScale Thermophoresis data. Binding parameters such as dissociation constants (Kd) or EC50 values and can be easily quantified and compared.
